Cryo-EM Facility

<p> <br /> </p> <p> A world class cryo-electron microscopy (Cryo-EM) facility is the foundation of Tsinghua University Branch of China National Center for Protein Sciences(Beijing). Cryo-EM facility of Tsinghua University provides expertise a nd resources for understanding both molecular a nd cellular structures by using cryo-EM and three-dimensional reconstruction tech nique. The facility has 8 tra nsmission electron microscopes,an FIB/SEM dual bea m system, a nd a correlative light-electron microscopy (CLEM) system, as well as a full set of cryo-sample preparation apparatuses including cryo-plungers, plasma clea ners a nd carbon evaporators. </p> <p> <br /> </p> <p> <br /> </p> <p style="color:#000000;font-family:&font-size:14px;font-style:normal;font-variant:normal;font-weight:400;letter-spacing:normal;margin-bottom:5px;margin-left:0px;margin-right:0px;margin-top:5px;orphans:2;overflow-wrap:break-word;text-align:left;text-decoration:none;text-indent:0px;text-transform:none;-webkit-text-stroke-width:0px;white-space:normal;word-spacing:0px;"> <br /> </p> <p> <p class="MsoNormal" style="text-indent:24.0pt;"> 冷冻电镜平台主要应用冷冻电镜三维重构技术,实现生物学样品的高分辨三维结构解析。研究对象主要包括生物大分子复合物、亚细胞器乃至细胞、组织等,通过多尺度的三维结构分析来解答生命科学的前沿问题。平台的主要仪器包括<span>10</span>台各类冷冻透射电子显微镜(含<span>300</span>千伏<span>Titan Krios 4</span>台)、<span>2</span>台聚焦离子束<span>/</span>扫描电子显微镜、冷冻光镜和一系列相关辅助制样设备。<span></span> </p> <p class="MsoNormal" style="text-indent:24.0pt;"> 本平台开展的服务项目包括镀膜、等离子清洗、负染样品制备、快速冷冻样品制备、高压冷冻、冷冻替代、冷冻超薄切片、透射电镜成像以及数据采集、扫描电镜成像以及数据收集、聚焦离子束加工,光镜电镜关联等,可实现单颗粒分析、电子断层成像分析和微晶电子衍射等方面的研究。<span></span> </p> <p class="MsoNormal" style="text-indent:24.0pt;"> 应用本平台仪器,一大批顶级成果,例如剪接体、离子通道、藻胆体、植物抗病蛋白、染色质重塑复合物、呼吸链复合物、新冠病毒等高分辨率的结构解析,引起了全世界的瞩目。本平台已经建设成为运行效率最高、最具国际知名度的冷冻电镜平台之一,是国内外众多已有或待建的冷冻电镜平台的标杆及模板。<span></span> </p> </p>
